Overview

IBI363 + fruquintinib is a combination therapy currently being evaluated in Phase 1 clinical trials for the treatment of advanced colorectal cancer, specifically targeting patients with microsatellite stable (MSS) or proficient mismatch repair (pMMR) tumors. IBI363 is a first-in-class bispecific antibody fusion protein that simultaneously blocks the Programmed Cell Death Protein 1 (PD-1) pathway and provides biased Interleukin-2 (IL-2) signaling. By targeting IL-2α (CD25), IBI363 is designed to selectively stimulate tumor-specific T cells while minimizing the systemic toxicity typically associated with high-dose IL-2 therapy. Fruquintinib (also referred to as furuitinib) is a highly selective and potent small molecule tyrosine kinase inhibitor that targets Vascular Endothelial Growth Factor Receptors (VEGFR) 1, 2, and 3, thereby inhibiting tumor angiogenesis. The combination is intended to convert immunologically "cold" tumors into "hot" tumors by combining immune checkpoint blockade and cytokine stimulation with anti-angiogenic effects.
