IBI377 is a recombinant humanized monoclonal antibody targeting interleukin-25 (IL-25), also known as IL-17E, developed by Innovent Biologics. IL-25 is a pro-inflammatory cytokine that plays a critical role in initiating and amplifying Type 2 immune responses, which are implicated in the pathogenesis of various inflammatory and autoimmune diseases. In the context of acute graft-versus-host disease (aGvHD), IL-25 levels are often elevated and correlate with disease severity, contributing to the inflammatory milieu that drives tissue damage. IBI377 binds to IL-25 with high affinity, neutralizing its biological activity and suppressing the downstream inflammatory cascade. This mechanism aims to provide a novel therapeutic approach for patients with aGvHD, particularly those who are refractory to or dependent on corticosteroids.
