Overview

IBI397 (also known as AL008) is a monoclonal antibody developed to target the SIRPα (Signal-regulatory protein alpha) pathway. It acts as a SIRPα inhibitor, disrupting the CD47-SIRPα interaction that cancer cells use to evade immune surveillance by sending a "don't eat me" signal to macrophages. By blocking this inhibitory receptor on macrophages and engaging Fc gamma receptors, IBI397 aims to enhance anti-tumor immunity through increased phagocytosis and immunostimulatory activity. The drug was initially developed by Alector and later licensed for development in China by Innovent Biologics for oncology indications. Clinical development reached Phase 1 in China for advanced cancers but has since been discontinued[1][2][3].
