Overview

IBIO-101 is a **second-generation anti-CD25 (IL2Rα) monoclonal antibody** developed by iBio to target immunosuppressive regulatory T (Treg) cells in the tumor microenvironment for oncology applications, especially solid tumors. The antibody is designed to **deplete Treg cells** via engagement of natural killer (NK) cells, promoting anti-tumor immunity by eliminating immunosuppressive elements. Unlike first-generation anti-CD25 antibodies, IBIO-101 is engineered to **avoid blocking interleukin-2 (IL-2) signaling to effector T cells (Teffs)**, thereby preserving anti-tumor immune responses. It has been developed using both traditional mammalian and plant-based *FastPharming* systems with iBio's AI-driven epitope steering and *Glycaneering* technologies. The drug is currently in preclinical development for solid tumors[3][4][7][8][9].
