Overview

IBR120 is a novel small molecule inhibitor of the RAD51 recombinase, a protein essential for homologous recombination repair (HRR) of DNA double-strand breaks. By disrupting RAD51 multimerization, IBR120 impairs the repair of DNA damage, leading to the induction of apoptotic cell death in cancer cells. It has demonstrated potent growth inhibition in triple-negative breast cancer (TNBC) cell lines and a broad spectrum of other cancer types, showing a significant therapeutic window with approximately 10-fold higher IC50 values in normal cells compared to cancer cells. IBR120 represents a potential therapeutic strategy for difficult-to-treat cancers that rely on RAD51-mediated DNA repair for survival and proliferation.
