Overview

Ibrutinib + obinutuzumab is a fixed-dose combination regimen of two distinct agents used for the treatment of chronic lymphocytic leukemia (CLL) and small lymphocytic lymphoma (SLL). Ibrutinib is an oral, first-in-class small molecule inhibitor of Bruton’s tyrosine kinase (BTK), a key enzyme in the B-cell receptor signaling pathway that promotes survival and proliferation of malignant B cells. Obinutuzumab is a fully humanized type II anti-CD20 monoclonal antibody that induces direct cell death and enhances antibody-dependent cellular cytotoxicity against CD20-expressing B cells. The combination provides a chemotherapy-free, targeted therapy option for treatment-naïve CLL/SLL patients, offering significant improvements in progression-free survival compared to traditional chemoimmunotherapy regimens[1][2][4].
