Overview

Ibrutinib + RICE is a combination therapy used in the treatment of relapsed or refractory mature B-cell non-Hodgkin lymphoma (B-NHL), particularly in pediatric and young adult patients. Ibrutinib is a small molecule inhibitor that irreversibly binds to Bruton's tyrosine kinase (BTK), blocking B-cell receptor signaling and inhibiting malignant B-cell proliferation and survival. The RICE regimen consists of rituximab (a monoclonal antibody targeting CD20 on B cells), ifosfamide (an alkylating agent), carboplatin (a platinum-based chemotherapeutic), and etoposide (a topoisomerase II inhibitor). This combination aims to enhance anti-tumor activity by integrating targeted BTK inhibition with established cytotoxic chemotherapy. Ibrutinib was developed by Pharmacyclics and is marketed as Imbruvica[1][4][5][6].
