Overview

This combination regimen consists of the non-steroidal anti-inflammatory drug (NSAID) ibuprofen, the granulocyte colony-stimulating factor (G-CSF) analog lenograstim (Myelostim), and the CXCR4 antagonist plerixafor (Mozobil). It is being investigated by IRCCS San Raffaele for the mobilization of hematopoietic stem cells (HSCs) in patients with X-linked Chronic Granulomatous Disease (X-CGD). In X-CGD, standard mobilization with G-CSF alone is often suboptimal or associated with inflammatory flares; the addition of ibuprofen is intended to enhance mobilization and potentially mitigate inflammation by inhibiting cyclooxygenase (COX) and reducing prostaglandin E2 (PGE2) levels, which are known to influence HSC retention in the bone marrow niche.
