Drug intelligence / Profile preview
This entry refers to a group of three analgesic and antipyretic agents—**ibuprofen**, **paracetamol** (acetaminophen), and **metamizole** (dipyrone)—that were evaluated by Mantecorp Indústria Química e Farmacêutica (now Hypera Pharma) in a Phase 4 post-marketing clinical trial (NCT01359020). The study was designed to compare the safety and efficacy of these small-molecule therapies in pediatric patients suffering from fever. Ibuprofen is a nonsteroidal anti-inflammatory drug (NSAID) that inhibits cyclooxygenase enzymes (COX-1 and COX-2). Paracetamol is a widely used analgesic and antipyretic that acts primarily through central mechanisms, including the inhibition of prostaglandin synthesis and modulation of the endocannabinoid system. Metamizole is a pyrazolone derivative with potent antipyretic, analgesic, and spasmolytic properties, acting via COX inhibition and potentially through central opioid and cannabinoid pathways. These drugs were studied as individual treatments within the trial rather than as a single fixed-dose combination product.
