Ibuprofen + tramadol is a fixed-dose combination analgesic being developed for the management of acute severe postoperative pain. This combination utilizes a multimodal approach to pain management, combining the anti-inflammatory and peripheral analgesic effects of ibuprofen with the centrally acting analgesic properties of tramadol. Tramadol acts as a mu-opioid receptor agonist and also inhibits the reuptake of serotonin and norepinephrine, enhancing descending inhibitory pain pathways. The specific formulation under development by Farmalider S.A. is an intravenous (IV) solution containing ibuprofen arginate (to enhance solubility) and tramadol hydrochloride, intended for use in adults undergoing surgery to provide effective analgesia while potentially reducing the overall requirement for higher-dose opioid monotherapy.
