Ibuprofen l-lysine is a water-soluble salt form of the nonsteroidal anti-inflammatory drug (NSAID) ibuprofen, created by combining racemic (±)-ibuprofen with the amino acid L-lysine. This formulation allows for faster absorption and is suitable for intravenous administration. Its primary indication is the closure of clinically significant patent ductus arteriosus (PDA) in premature infants weighing between 500 and 1500 grams who are no more than 32 weeks gestational age when usual medical management has failed[1][2][3]. The mechanism of action involves inhibition of cyclooxygenase (COX), leading to reduced synthesis of prostaglandins that mediate inflammation, pain, fever, and maintenance of ductal patency in neonates[6]. By reducing prostaglandin levels, it promotes constriction and closure of the PDA. Ibuprofen l-lysine may also be used orally for short-term symptomatic relief of headache and migraine in adults and adolescents[8].
