IC135 is an experimental immunocytokine designed for the treatment of acute myeloid leukemia (AML). It consists of an Fc-optimized monoclonal antibody targeting CD135 (also known as FLT3) fused to wild-type interleukin-15 (IL-15). The construct is intended to concentrate IL-15 activity at the site of leukemic cells, thereby stimulating natural killer (NK) cell proliferation and enhancing antibody-dependent cellular cytotoxicity (ADCC) against the tumor. While IC135 demonstrates potent anti-leukemic activity in preclinical models, its use of wild-type IL-15 allows for binding to the IL-15 receptor alpha (IL-15Rα) on healthy cells, which can lead to non-specific immune activation and off-target toxicity. This profile led to the development of modified versions, such as MIC135, which aim to restrict IL-15 activity solely to the target-cell interface.
