Overview

iC9-CAR.CSPG4 T cells are an investigational autologous chimeric antigen receptor (CAR) T cell therapy engineered to express both a CAR targeting the tumor-associated antigen chondroitin sulfate proteoglycan 4 (CSPG4) and an inducible caspase 9 (iC9) safety switch[3][7][9]. The therapy is designed to recognize and eliminate CSPG4-expressing tumor cells—CSPG4 is upregulated in a range of solid tumors, including melanoma, glioblastoma, triple-negative breast cancer, and head and neck squamous cell carcinoma (HNSCC)[6][8][10]. The inducible caspase 9 suicide gene provides a safety mechanism to selectively ablate the engineered T cells if severe toxicity occurs. The primary indication under current clinical investigation is relapsed or refractory head and neck squamous cell carcinoma[1][9]. iC9-CAR.CSPG4 T cells are being developed in academic settings and in early-phase clinical trials; neither a specific commercial developer nor manufacturer is specified[1][7][9].
