iC9.ADR.CD19.CAR.NKT cells are an experimental chimeric antigen receptor (CAR)–engineered invariant natural killer T (iNKT) cell therapy directed against the B‑cell antigen CD19 and incorporating an inducible caspase 9 (iC9) suicide switch and an additional ADR safety or selection element. Built on the CAR‑NKT platform, these allogeneic iNKT cells are genetically modified to express a CD19‑specific CAR for targeted killing of CD19‑positive B‑cell malignancies and to allow pharmacologic elimination of the cells via iC9 activation in case of severe toxicity, while retaining the endogenous iNKT T‑cell receptor and NK‑like effector functions. Preclinical CD19 CAR‑NKT models have demonstrated potent anti‑tumor activity with prolonged in vivo persistence when using central‑memory–like iNKT subsets, supporting development of this construct as an off‑the‑shelf cell therapy for CD19‑expressing hematologic cancers.[5]
