Overview

ICA-1 is a small molecule inhibitor of protein kinase C-iota (PKC-ι) currently in preclinical development for the treatment of various cancers. It specifically targets a unique amino acid sequence (residues 469-475) within the catalytic domain of PKC-ι, an oncogenic protein kinase that is frequently overexpressed in solid tumors and plays a critical role in cell survival and proliferation. Developed by Suzhou Aikailiyuan Biotechnology, ICA-1 has demonstrated the ability to inhibit cell proliferation and induce apoptosis in neuroblastoma models. Beyond neuroblastoma, research suggests potential applications in treating other solid tumors, including prostate, breast, ovarian, melanoma, and glioma. A related dephosphorylated analog, ICA-1S, has also been studied for its toxicological profile and potential for oral administration.
