Drug intelligence / Profile preview
ICA-264 is a novel, subnanomolar, orally active small molecule apolipoprotein L1 (ApoL1) inhibitor developed by OmniAb in collaboration with BioPharmaWorks. It is designed to target the ion channel function of ApoL1, specifically inhibiting both the ApoL1-G1 and ApoL1-G2 genetic variants, which are associated with an increased risk of chronic kidney disease (CKD) and other nephropathies in individuals of African ancestry. In preclinical studies, ICA-264 demonstrated potent, concentration-dependent inhibition of ApoL1-mediated thallium (Tl+) flux and directly blocked ApoL1 currents in electrophysiological assays. In vivo, oral administration of ICA-264 significantly reduced proteinuria and decreased the urinary albumin-to-creatinine ratio (uACR) in transgenic mouse models of ApoL1-induced nephropathy, highlighting its potential as a therapeutic agent for glomerular diseases.
