ICAM-2 shRNA is a short hairpin RNA-based experimental agent designed to silence the expression of Intercellular Adhesion Molecule 2 (ICAM-2/CD102). In the context of acute myeloid leukemia (AML), ICAM-2 is highly expressed on leukemia blasts and mediates their adhesion to the bone marrow microenvironment (BMME), particularly bone marrow stromal cells. This adhesion facilitates chemoresistance by activating pro-survival signaling pathways, including PI3K/AKT/mTOR and NF-κB, and upregulating anti-apoptotic proteins such as Bcl-2 and MCL-1. By utilizing a lentiviral vector to deliver shRNA, researchers can effectively knock down ICAM-2 expression, thereby disrupting the protective anchoring of AML cells to the BMME and sensitizing them to chemotherapy agents like cytarabine and daunorubicin.
