ICAM1 ADC 1 is an experimental antibody-drug conjugate (ADC) targeting Intercellular Adhesion Molecule 1 (ICAM1), currently in preclinical development for the treatment of endometrial cancer. Developed by researchers at the Hangzhou Institute of Medicine (HIM), Chinese Academy of Sciences, and Zhejiang Cancer Hospital, this agent is one of two ICAM1-targeted ADCs described in preclinical studies presented at AACR 2024. The drug consists of a monoclonal antibody specific to ICAM1 conjugated to a cytotoxic payload via a clinically-validated linker. ICAM1 is significantly overexpressed in endometrial cancer tissues compared to normal paracancerous tissues, and the ADC is designed to selectively bind these cells and undergo antigen-mediated endocytosis to deliver its payload. In preclinical cell line-derived xenograft (CDX) models, ICAM1 ADCs demonstrated potent anti-tumor activity and a favorable safety profile, outperforming standard-of-care chemotherapy.
