ICAM1 ADC 2 is an experimental antibody-drug conjugate (ADC) designed to target Intercellular Adhesion Molecule 1 (ICAM1), a cell surface glycoprotein that is significantly overexpressed in endometrial cancer tissues compared to normal paracancerous tissues. Developed by researchers at the Hangzhou Institute of Medicine and Zhejiang Cancer Hospital, the drug utilizes a clinically-validated linker and payload combination conjugated to an anti-ICAM1 antibody. The mechanism involves selective recognition of ICAM1 on tumor cells followed by antigen-mediated endocytosis, which delivers the cytotoxic payload directly into the cancer cells. In preclinical studies presented at AACR 2024, ICAM1 ADC 2 demonstrated potent anti-tumor activity in cell line-derived xenograft (CDX) models of endometrial cancer, outperforming standard-of-care chemotherapy without observed toxicity.
