ICAM1-DXd is an antibody-drug conjugate (ADC) composed of a monoclonal antibody targeting Intercellular Adhesion Molecule 1 (ICAM1) conjugated to the topoisomerase I inhibitor payload, DXd (deruxtecan), via a cathepsin B-cleavable tetrapeptide-based linker. ICAM1 is a cell surface glycoprotein often overexpressed in various malignancies, including cervical cancer. Beyond its direct cytotoxic effect on ICAM1-expressing tumor cells, ICAM1-DXd has been shown to non-specifically eliminate pro-tumor tumor-associated neutrophils (TANs) through phagocytosis. This depletion of immunosuppressive TANs within the tumor microenvironment enhances the infiltration and activity of CD8+ T cells, providing a mechanistic basis for its synergistic anti-tumor activity when combined with PD-1 checkpoint inhibitors.
