ICAM1-IRDye700 is an experimental theranostic conjugate designed for near-infrared photoimmunotherapy (NIR-PIT) and fluorescence-guided imaging. It consists of a monoclonal antibody targeting Intercellular Adhesion Molecule 1 (ICAM-1/CD54) covalently linked to the near-infrared phthalocyanine dye IRDye 700DX. ICAM-1 is a cell surface glycoprotein that is frequently overexpressed in various malignancies, including triple-negative breast cancer (TNBC) and head and neck squamous cell carcinoma, as well as in inflammatory environments. The mechanism of action involves the selective binding of the conjugate to ICAM-1-expressing cells; upon subsequent exposure to 690 nm near-infrared light, the IRDye 700DX moiety undergoes a photochemical reaction that induces rapid physical damage to the cell membrane, leading to immunogenic cell death. Beyond its therapeutic application, the fluorescent properties of the IRDye 700DX payload allow for real-time visualization of tumor margins and monitoring of therapeutic efficacy.
