Overview

iCAR-ILC-N101 is an investigational, off-the-shelf cell therapy composed of innate lymphoid cells (ILC) or natural killer (NK) cells derived from induced pluripotent stem cells (iPSCs). These cells are genetically engineered to express a chimeric antigen receptor (CAR) specific for Glypican-3 (GPC3), a cell-surface protein frequently overexpressed in certain solid tumors, including ovarian clear cell carcinoma. Developed by researchers at Kyoto University with support from the Japan Agency for Medical Research and Development (AMED), iCAR-ILC-N101 is currently being evaluated in a Phase 1 clinical trial for patients with GPC3-positive advanced ovarian clear cell carcinoma (OCCC) and peritoneal dissemination. The therapy is administered via intraperitoneal injection to maximize local exposure to tumor cells within the peritoneal cavity, leveraging the innate cytotoxic potential of ILC/NK cells combined with the targeted specificity of the CAR construct.
