Overview

IcasM28z CAR T cells is an autologous, second-generation chimeric antigen receptor (CAR) T-cell therapy targeting mesothelin (MSLN), developed by Memorial Sloan Kettering Cancer Center in collaboration with Bellicum Pharmaceuticals. The CAR construct consists of a fully human mesothelin-specific single-chain variable fragment (scFv) linked to CD28 and CD3-zeta signaling domains. It is engineered with an inducible caspase-9 (iCasp9) safety 'suicide' switch, which can be activated by a dimerizing agent (such as rimiducid) to induce apoptosis of the CAR T cells in the event of severe on-target, off-tumor toxicity. The therapy is primarily investigated for the treatment of mesothelin-expressing solid tumors, including malignant pleural mesothelioma, non-small cell lung cancer, and breast cancer.
