Overview

iCasp9M28z T cells are an autologous chimeric antigen receptor (CAR) T cell therapy engineered to target mesothelin, a protein highly expressed on the surface of several solid tumors, including malignant pleural mesothelioma and other cancers. These genetically modified T cells express a CAR composed of an anti-mesothelin single-chain variable fragment (scFv), CD28 and CD3ζ signaling domains, and incorporate an inducible caspase 9 (iCasp9) suicide switch for safety. Upon administration, these cells specifically recognize and kill mesothelin-expressing tumor cells through immunologic cytotoxicity. The inclusion of the iCasp9 safety switch allows for selective elimination of the infused CAR-T cells in case of severe adverse events. This therapy is being developed primarily by Atara Biotherapeutics under license from Memorial Sloan Kettering Cancer Center[1][2][3][5].
