ICBN-PDL1 is an immune checkpoint-blocking nanocage designed to overcome the limitations of monoclonal antibodies in treating brain tumors. It consists of a 24-subunit human ferritin heavy-chain (HFn) nanocage platform that displays 24 copies of a PD-L1-binding peptide (specifically the sequence CVRARTR) on its surface. This biologic modality leverages the natural ability of ferritin to cross the blood-brain barrier (BBB) via transferrin receptor 1 (TfR1) mediated transcytosis. By targeting Programmed Death-Ligand 1 (PD-L1), ICBN-PDL1 disrupts the PD-1/PD-L1 inhibitory axis, thereby restoring T-cell activity within the immunosuppressive microenvironment of tumors like glioblastoma. Preclinical studies have demonstrated its efficacy in both orthotopic intracranial and allograft colon cancer models.
