ICG-gemcitabine (ICG-Gem) is an investigational small molecule conjugate designed as a liver-specific fluorescent drug delivery system for the treatment and diagnosis of hepatocellular carcinoma (HCC). It consists of the FDA-approved tricarbocyanine dye indocyanine green (ICG) covalently conjugated to the chemotherapeutic agent gemcitabine. ICG serves as a targeting moiety that specifically accumulates in hepatocytes and HCC cells, while gemcitabine acts as the cytotoxic payload. Preclinical studies indicate that ICG-gemcitabine exhibits reduced toxicity to normal cells and enhanced anti-tumor efficacy compared to gemcitabine alone. Its fluorescent properties also allow for potential use in image-guided therapy or diagnosis of liver cancer.
