ICG-POL6326 is a near-infrared-II (NIR-II) fluorescent molecular imaging probe developed for the intraoperative visualization of breast cancer tissue and metastatic sentinel lymph nodes (SLNs). It is synthesized by covalently conjugating the near-infrared dye indocyanine green (ICG) with POL6326 (balixafortide), a potent and selective peptide antagonist of the CXC chemokine receptor 4 (CXCR4). Since CXCR4 is highly expressed in metastatic breast cancer cells, the probe specifically targets and accumulates in metastatic SLNs and tumor tissues. This allows real-time, high-contrast, and deep-penetration fluorescence imaging during breast-conserving surgery, potentially aiding in the rapid intraoperative detection of lymph node metastasis and reducing the need for axillary lymph node dissection.
