**ICG144** is an investigational, lentivirally engineered compound chimeric antigen receptor T-cell therapy for relapsed or refractory high-risk hematologic malignancies, principally acute myeloid leukemia. The product expresses two complete CAR constructs linked by a self-cleaving P2A peptide: an anti-CLL1 CAR designed to address leukemia stem cell populations and an anti-CD33 CAR directed at bulk AML blasts. The CLL1 CAR incorporates CD28 costimulation, while the CD33 CAR incorporates 4-1BB costimulation; both use CD3ζ signaling. The construct also includes a CD52-based safety switch intended to permit pharmacologic elimination of infused CAR-T cells with alemtuzumab if required.
