Overview

ICG415 is an investigational autologous chimeric antigen receptor T-cell (CAR-T) therapy developed by iCell Gene Therapeutics for the treatment of relapsed or refractory acute myeloid leukemia (AML). It utilizes a compound CAR (cCAR) design that simultaneously targets two distinct antigens: CD33, which is expressed on the majority of AML blasts, and CLL1 (C-type lectin-like molecule-1), which is highly expressed on AML cells and leukemia stem cells (LSCs) but absent on healthy hematopoietic stem cells. This dual-targeting approach is intended to enhance anti-leukemic efficacy and prevent antigen escape, a common mechanism of resistance and relapse in single-target CAR-T therapies. ICG415 is currently being evaluated in Phase I clinical trials to assess its safety and preliminary efficacy in adult patients.
