ICON-1 is a first-in-class recombinant fusion protein developed by Iconic Therapeutics to target Tissue Factor (TF), a protein overexpressed in various cancers and pathological neovascularization. The molecule is composed of a catalytically inactive human Factor VIIa (FVIIa) domain fused to the Fc region of a human IgG1 antibody. By utilizing the high affinity of FVIIa for TF, ICON-1 acts as an immunoconjugate that binds to TF-expressing tumor cells and pathological blood vessels. The IgG1 Fc domain then recruits immune effector cells, such as natural killer (NK) cells, to induce antibody-dependent cell-mediated cytotoxicity (ADCC), leading to the destruction of the target cells. In uveal melanoma, ICON-1 has been investigated via intravitreal injection to evaluate its safety and ability to target ocular tumor cells in patients scheduled for surgical interventions.
