This is a combination therapy consisting of icotinib, a first-generation epidermal growth factor receptor tyrosine kinase inhibitor (EGFR-TKI), and rapamycin, an mTOR inhibitor also known as sirolimus. Icotinib selectively inhibits the ATP binding site of EGFR, blocking downstream signaling pathways that drive cell proliferation in cancers with EGFR mutations. Rapamycin inhibits the mammalian target of rapamycin (mTOR), a key regulator of cell growth and proliferation. The combination has been explored in case reports for advanced non-small cell lung cancer (NSCLC) in post-transplant patients to leverage potential synergistic antitumor effects by targeting both EGFR and mTOR pathways. There are no approved indications or large clinical trials for this specific combination; its use is experimental and primarily reported in individual cases[1][2][3].
