Icovamenib is an investigational, orally bioavailable, potent, and selective covalent inhibitor of menin. Developed using Biomea Fusion’s FUSION™ System, it is designed to regenerate insulin-producing pancreatic beta cells with the aim of curing diabetes. Icovamenib’s mechanism of action involves enabling the proliferation, preservation, and reactivation of a patient’s own healthy functional beta cells. Clinical studies have shown that treatment with icovamenib leads to significant improvements in pancreatic beta-cell function (as measured by increased C-peptide levels) and durable reductions in HbA1c in patients with severe insulin-deficient diabetes. The drug has demonstrated benefits both as monotherapy and in combination with Glucagon-like peptide 1 (GLP-1)-based therapies such as semaglutide. Icovamenib is being developed as a potentially first-in-class disease-modifying therapy for both type 1 and type 2 diabetes.
