ICP-054 is a potent and selective small molecule antagonist of the C-C chemokine receptor type 8 (CCR8), currently under development by InnoCare Pharma for the treatment of advanced solid tumors. CCR8 is highly and specifically expressed on tumor-infiltrating regulatory T cells (Tregs), which play a critical role in maintaining an immunosuppressive tumor microenvironment. By antagonizing CCR8, ICP-054 aims to selectively inhibit the recruitment and suppressive function of these Tregs within the tumor, thereby enhancing the anti-tumor immune response mediated by effector T cells. Unlike many other Treg-targeting strategies, ICP-054's selectivity for tumor-infiltrating Tregs over peripheral T cell populations potentially reduces the risk of systemic immune-related adverse events. The drug is being investigated as both a monotherapy and in combination with other immunotherapies, such as PD-1/PD-L1 inhibitors, to overcome resistance in various cancer types.
