ICP-189 is a potent and selective oral allosteric inhibitor of SHP2 (Src Homology 2 domain containing protein tyrosine phosphatase), developed by InnoCare for the treatment of solid tumors, including non-small cell lung cancer (NSCLC). It acts by inhibiting SHP2, a key mediator in the MAPK signaling pathway and immune checkpoint pathways, thereby regulating cellular proliferation and survival. Preclinical studies have demonstrated its excellent selectivity over other phosphatases and significant anti-tumor effects in various xenograft models. ICP-189 is being evaluated both as monotherapy and in combination with other targeted or immuno-oncology agents such as EGFR inhibitors (e.g., furmonertinib), MEK inhibitors, KRASG12C inhibitors, and anti-PD-1 antibodies. The drug has shown promising preliminary efficacy with a favorable safety profile in early clinical trials[1][3][7][10].
