ICP-488 is an oral, potent, and selective allosteric inhibitor of tyrosine kinase 2 (TYK2), developed by InnoCare Pharma for the treatment of autoimmune and inflammatory diseases, primarily psoriasis. It acts by binding to the Janus Homology 2 (JH2) pseudokinase domain of TYK2, thereby blocking signal transduction from cytokines such as IL-23, IL-12, and type I interferons. This inhibition disrupts key pathological processes in autoimmune conditions. Clinical trials have shown that ICP-488 achieves significant improvements in Psoriasis Area and Severity Index (PASI) scores with a favorable safety profile; most adverse events are mild or moderate. The drug is currently in phase III clinical development for plaque psoriasis in China[1][3][4][7].
