ICP-490 is an orally available, next-generation small molecule immunomodulatory imide drug (IMiD) developed by InnoCare Pharma for the treatment of multiple myeloma and non-Hodgkin's lymphoma. It functions as a molecular glue degrader, specifically binding to the CRL4CRBN-E3 ligase complex to induce ubiquitination and degradation of transcription factors IKZF1 (Ikaros) and IKZF3 (Aiolos), which are critical for tumor cell survival in hematologic malignancies. This mechanism allows ICP-490 to overcome resistance seen with earlier-generation cereblon modulators such as lenalidomide. Preclinical studies have demonstrated potent anti-tumor activity against multiple myeloma and non-Hodgkin’s lymphoma, including diffuse large B-cell lymphoma (DLBCL), with minimal cytotoxicity toward normal cells. The drug is currently in phase 2 clinical trials for relapsed/refractory multiple myeloma and non-Hodgkin's lymphoma[1][2][4][5][6].
