ICP-538 is a novel, potent, and highly selective molecular glue degrader designed for oral administration. It targets Vav guanine nucleotide exchange factor 1 (VAV1), a critical signaling protein located downstream of T-cell and B-cell receptors that plays a key role in immune cell activation. By inducing the degradation of VAV1, ICP-538 aims to modulate aberrant immune responses associated with various autoimmune conditions. Developed by InnoCare Pharma, the compound received Investigational New Drug (IND) approval from the China National Medical Products Administration (NMPA) in February 2026, marking it as the first VAV1 degrader to enter clinical trials in China. Its primary therapeutic focus includes inflammatory bowel disease (IBD), systemic lupus erythematosus (SLE), and multiple sclerosis (MS).
