Overview

iCP-Parkin is a recombinant, cell- and tissue-permeable form of the human Parkin protein engineered for enhanced intracellular delivery and solubility. Developed by Cellivery, it is designed to cross the blood-brain barrier and deliver functional Parkin directly into neurons. The drug’s mechanism of action involves mimicking endogenous Parkin (an E3 ubiquitin ligase), promoting mitophagy (the selective removal of damaged mitochondria), enhancing mitochondrial biogenesis, and facilitating the clearance of toxic α-synuclein aggregates via both the ubiquitin-proteasome system and autophagy-lysosome pathway. Preclinical studies in cellular and animal models of Parkinson’s disease have shown that iCP-Parkin can restore mitochondrial function, reduce α-synuclein accumulation, protect dopaminergic neurons from degeneration, improve motor symptoms after onset, and potentially reverse disease progression. It is being developed as a first-in-class disease-modifying therapy for Parkinson’s disease[1][2][3][4][5][6][7].
