Overview

Icrucumab is a fully human IgG1 monoclonal antibody that targets and binds with high specificity to vascular endothelial growth factor receptor 1 (VEGFR-1, also known as Flt-1). By blocking the interaction between VEGFR-1 and its proangiogenic ligands (including VEGF-A, VEGF-B, and PlGF), icrucumab inhibits downstream signaling pathways involved in tumor angiogenesis. This results in reduced blood vessel formation within tumors, limiting their oxygen and nutrient supply, thereby inhibiting tumor cell proliferation and metastasis. Icrucumab was developed for the treatment of solid tumors such as breast cancer, colorectal cancer, and urogenital cancers. Its development was led by ImClone Systems (later acquired by Eli Lilly), but clinical progress has been discontinued after phase II trials due to limited efficacy[3][4][5][6][8].
