Overview

ICT41 is a humanized, anti-BTN3A monoclonal antibody developed by ImCheck Therapeutics. It selectively activates γ9Vδ2 T cells, which are key components of the immune system's first line of defense against infectious diseases. By binding with high affinity to all three BTN3A isoforms, ICT41 induces a conformational change in these proteins that enables engagement with the γ9Vδ2 T cell receptor (TCR). This leads to rapid activation and migration of γ9Vδ2 T cells from blood to infected tissues, triggering pro-inflammatory cytokine release (such as IFNγ and TNFα) and activating other immune cells including NK cells, CD8+ T cells, dendritic cells, and B cells. Activated γ9Vδ2 T cells can directly kill infected target cells via cytotoxic granule release (perforin/granzymes/granulysins), death receptor pathways, and antibody-dependent cellular cytotoxicity (ADCC). The drug is being developed primarily for pathogen-agnostic treatment of viral and bacterial infections[1][4][5].
