Overview

ICVB-1042 is a chimeric oncolytic adenovirus rationally engineered for tumor-selective replication and enhanced tumor cell killing. It incorporates specific genomic modifications, including E2F-dependent elements and a chimeric fiber, to increase its selectivity for cancer cells overexpressing CD46 and to broaden its tropism across various solid tumors. The virus is designed for both intravenous and intratumoral administration, with preclinical studies demonstrating high potency against multiple tumor types (including head and neck, bladder, lung, and breast cancers), robust immune activation, reduced off-target toxicity in normal tissues, and effective systemic delivery. Its mechanism involves selective infection of cancer cells via CD46 binding followed by viral replication leading to tumor cell lysis[1][5][6][7]. Developed by IconOVir Bio (now acquired by UroGen), ICVB-1042 is currently in Phase 1 clinical trials for advanced malignant solid neoplasms[3][4].
