ICX-101 is an investigational small molecule MYC inhibitor developed by Incurix. It functions by directly disrupting the interaction between the MYC/MAX heterodimer and its target DNA binding sequence, the E-box. By preventing this protein-DNA interaction, ICX-101 inhibits the transcriptional activity of MYC, a transcription factor frequently dysregulated in various malignancies. Preclinical studies have demonstrated antitumor activity in patient-derived cells of advanced lung cancer, particularly in KRAS-mutant non-small cell lung cancer (NSCLC) and small cell lung cancer (SCLC), with efficacy correlating with MYC expression levels. The drug is currently in preclinical development, with GLP studies indicated in company pipeline reports.
