ICX-201

ICX-201 is a preclinical small molecule inhibitor of Signal transducer and activator of transcription 3 (STAT3), being developed by Incurix for the treatment of STAT3-driven cancers. The compound is designed to selectively inhibit STAT3 by interfering with the biophysical binding between the STAT3 protein and DNA, specifically targeting STAT3-dependent transcriptional activity and protein–DNA interactions. This mechanism differs from traditional SH2 domain inhibitors by directly blocking the interface between the transcription factor and its target DNA sequences, an approach Incurix utilizes to target transcription factors that are typically considered 'undruggable'.
