ID22 peptide vaccine is a synthetic peptide-based cancer vaccine developed for preclinical use. It consists of an 11-mer minimal neoantigen idiotype epitope fused via a four-amino acid linker to a heat shock cognate protein 70 (HSC70) binding motif. The vaccine is designed to enhance immunogenicity and elicit CD4 T cell-mediated immune responses when formulated with the poly(I:C) adjuvant. Upon subcutaneous administration in mice, ID22 induced strong antigen-specific CD4 T cell activation, as measured by IFN-γ production, and provided protective immunity against tumor challenge in a mouse model of mineral oil-induced plasmacytoma (MOPC.315.BM)[1]. The immune response was specific for the mutated idiotype epitope and not observed with wild-type control peptides containing the HSC70 binding motif. The developers synthesized ID22 and evaluated its efficacy and immunogenicity in preclinical murine studies. The mechanism of action centers on neoantigen-specific activation of CD4 T cells targeting the idiotype peptide sequence, with enhanced immunogenicity due to the HSC70 binding motif[1].
