**Id3-CAR T cells** are genetically engineered T cells overexpressing the transcription factor **Id3** to enhance anti-tumor efficacy and prevent exhaustion. Id3 modulates gene expression to sustain T cell proliferation, memory formation, and effector function during chronic antigen exposure, reducing exhaustion markers and promoting an NK-like phenotype with sustained expression of proliferation and memory genes. Developed through preclinical engineering, these cells demonstrate superior tumor control in mouse models of leukemia and lymphoma compared to standard CAR T cells, positioning them as an advanced cell therapy to overcome limitations in persistence and durability for hematological malignancies.
