This is a combination therapy consisting of three agents: - **Idasanutlin**: a small molecule MDM2 antagonist that disrupts the interaction between MDM2 and p53, leading to activation of the p53 pathway and apoptosis in cancer cells with wild-type p53. - **Venetoclax**: a small molecule BCL-2 inhibitor that triggers apoptosis in malignant lymphocytes by inhibiting BCL-2, an anti-apoptotic protein. - **Obinutuzumab**: a type II glycoengineered anti-CD20 monoclonal antibody that elicits B cell lysis through direct cytotoxicity and immune effector mechanisms. This regimen is being investigated for use in patients with relapsed or refractory follicular lymphoma (FL). The therapeutic hypothesis is that dual pathway targeting of apoptosis (MDM2-p53 and BCL2) combined with anti-CD20-mediated B cell depletion may enhance efficacy over single-agent or doublet regimens. The combination is currently under clinical evaluation in early-phase studies for relapsed/refractory follicular lymphoma[1].
