IDB-1829 is an orally bioavailable molecular glue degrader (MGD) developed by Interdict Bio for the treatment of metastatic castration-resistant prostate cancer (mCRPC). It is designed to target the androgen receptor (AR), a critical driver of prostate cancer progression, and induce its proteasomal degradation. By acting as a molecular glue, IDB-1829 facilitates a novel interaction between the AR and an E3 ubiquitin ligase, leading to the ubiquitination and subsequent destruction of the receptor protein. This mechanism is intended to overcome resistance to traditional AR signaling inhibitors, such as enzalutamide, which often occurs through AR amplification, mutations, or the expression of splice variants like AR-V7. IDB-1829 is currently being evaluated in a Phase 2a first-in-human clinical trial in Australia, both as a monotherapy and in combination with enzalutamide.
