IDE705 is a novel small molecule inhibitor targeting the helicase activity of DNA polymerase theta (Pol θ), also known as POLQ. It is being developed as a potential first-in-class therapy for solid tumors with homologous recombination deficiency (HRD). The drug is primarily investigated in combination with niraparib (a PARP inhibitor) to exploit synthetic lethality in HRD-positive cancers. IDE705 is under joint development by IDEAYA Biosciences and GSK and has entered Phase 1/2 clinical trials for advanced solid tumors[3][6][7]. Its mechanism involves inhibiting Pol θ-mediated DNA repair pathways that are critical for the survival of HR-deficient tumor cells.
