Drug intelligence / Profile preview
IDE892 + RG6620 (GDC-7035) is an investigational combination therapy being evaluated for the treatment of pancreatic ductal adenocarcinoma (PDAC) harboring both a methylthioadenosine phosphorylase (MTAP) deletion and a KRAS G12D mutation. IDE892 is a highly selective, methylthioadenosine (MTA)-cooperative inhibitor of protein arginine methyltransferase 5 (PRMT5) developed by IDEAYA Biosciences. It selectively binds to PRMT5 in the presence of MTA, exploiting the synthetic lethal vulnerability created by MTAP deletion in tumor cells. RG6620 (GDC-7035), developed by Genentech (a member of the Roche Group), is a selective small molecule inhibitor of the KRAS G12D mutant protein. By combining PRMT5 inhibition with KRAS G12D pathway blockade, this therapeutic regimen aims to deliver deeper and more durable anti-tumor responses in a molecularly defined patient population that currently lacks approved targeted treatment options.
