Overview

IdeXork (Xork) is a next-generation immunoglobulin G (IgG) protease designed to mitigate pre-existing anti-adeno-associated virus (AAV) antibodies in patients undergoing gene therapy. By cleaving systemic IgG, IdeXork aims to prevent the neutralization of AAV vectors, thereby expanding the eligible patient population for treatments such as Astellas' AT845 for Late-Onset Pompe disease (LOPD). The technology was originally licensed from Genovis by Selecta Biosciences (now Cartesian Therapeutics) and subsequently sublicensed to Astellas Pharma. However, Astellas terminated the development agreement effective June 2024, and Cartesian subsequently terminated its underlying license with Genovis in September 2024, effectively ending the program's development for Pompe disease.
